Oseltamivirphosphate is a neuraminidase inhibitor anti-influenza drug marketed in 1999 under the trade name Tamiflu(Tamiflu). It was first discovered by GileadSciences, then developed in partnership with Roche and approved for sale in the United States. Oseltamivir phosphate is the first orally effective neuraminidase inhibitor that is effective against both influenza A and B viruses. Oral oseltamivir is rapidly catalyzed by liver and intestinal esterase to its active metabolite, olchemicalbook seltamivir carboxylate. Oseltamivir carboxylate has a configuration similar to the transition state of neuraminic acid and binds competitively to the active site of influenza virus neuraminidase (NA). It is therefore a potent and highly selective influenza virus NA inhibitor (NAIs), which reduces the transmission of influenza A or B viruses mainly by interfering with the release of viruses from infected host cells. Therefore, oseltamivir phosphate is a precursor drug. At present, oseltamivir phosphate (Tamiflu) is still recognized as one of the most effective drugs on the market for the treatment of influenza A (H1N1), which was approved for market in China in July 2004.
Oseltamivide phosphate is rapidly metabolized to oseltamivir carboxylate by absorption through the gastrointestinal tract and at least 75 % of the oral dose is involved in systemic circulation in the form of oseltamivircarboxylate .The active ingredient Is a potent and unique neuraminidase inhibitor that acts on all aspects of influenza virus infection and prevents the replication of all clinically relevant strains of influenza A or B viruses.In vitro in very low nanomolarconcentrations that have inhibitory effect.
